When I issue an new assignment I would like to create a new custom field (say assignment_2) and populate it with the new filename
Can I populate it in xls and then just import AND UPDATE existing members?

When you import contacts into the database and that contact already exists then the imported data replaces the existing data, i.e. so after importing all imported contacts will have the assignment as specified in the import file.
